Biopharmaceutics Classification System (BCS) 

The BCS (Biopharmaceutics Classification System) was introduced in 1995 by Amidon et al., aiming to categorize drugs based on their solubility and permeability. The BCS was developed to help predict the in vivo behavior of pharmaceutical compounds, especially their absorption from oral dosage forms. 

The primary goal of the BCS classification is to streamline drug development and regulatory processes by categorizing drugs based on two key factors:

  • Solubility
  • Permeability

BCS Classification

The BCS classification divides drugs into four categories based on their solubility and permeability properties:

  • Class I: High Solubility, High Permeability

    • Drugs in this category dissolve well in water and can easily pass through biological membranes. 
    • These drugs typically exhibit good absorption and can be administered via conventional oral dosage forms like tablets or capsules.
  • Class II: Low Solubility, High Permeability

    • These drugs have poor solubility in water but are absorbed well through biological membranes once they are dissolved. 
    • Formulations need to be designed to improve solubility (e.g., through the use of solubilizing agents or nanoparticles) to achieve sufficient bioavailability.
  • Class III: High Solubility, Low Permeability

    • These drugs dissolve well in water but have limited ability to permeate biological membranes.
    • For these drugs, enhancing permeability is the primary focus of formulation development (e.g., using permeability enhancers or new delivery systems).
  • Class IV: Low Solubility, Low Permeability

    • Drugs in this class have both poor solubility and low permeability.
    • These are the most challenging drugs to develop for oral administration and may require advanced formulations such as lipid-based systems, microspheres, or drug delivery technologies to enhance both solubility and permeability.
